Chromatographic columns are a key process for successfully separating and purifying substances. It is crucial to monitor the separation effect and separation endpoint of each molecule during initial development and production. Conventional offline chromatographic testing is slow, resulting in a longer development cycle for purification processes.
Raman online can continuously analyze the composition of the mobile phase, monitor the concentration of separated components in real-time, quickly determine the separation endpoint, and help understand the impact of various process parameters on product quality and process efficiency;
The following figure shows the concentration of two molecular mixtures as they pass through the chromatographic column at the output end over time. It can be seen that the concentration of molecule 1 starts to increase from the 8th minute, reaches its maximum at 19 minutes, and drops to zero at 30 minutes, indicating that all molecule 1 has left the chromatographic column. The concentration of molecule 2 starts to increase from the 15th minute, reaches its highest point around 30 minutes, and decreases to zero at 45 minutes, ending the separation experiment. The resolution of this chromatographic column for separating two molecules is approximately 0.4.
